  1. What is the purpose of this lab?
  2. Which two major molecular tools have been used to create the recombined plasmid?
  3. What were the two sources of DNA used to create the recombined plasmid?
  4. You will set up four microtubes, each with different contents.  What will you place in microtube 1? 2? 3? 4?  Why?
  5. After preparing the four samples, you’ll place the microtubes in a 37 degrees Celsius water bath for 60 minutes.   What is the purpose of incubating the samples?  Why use a 37 degrees Celsius water bath?
  6. Study the restriction maps on page 5 of the lab.  Predict the results of lanes 1 and 2 on a gel.  In other words, how many bands do you expect to observe in lanes 1 and 2?  What is the size of each fragment?
