1.  What is the purpose of the Bradford assay?  How will it be used in this lab?

2.  How will you determine the amount of protein in each milk sample?

3.  The Bradford Dye Reagent is brown.  What happens when the reagent is placed in the presence of protein?

4.  Observe the standards.  What does the intensity of blue indicate in terms of relative amounts of protein?  In other words, does a dark blue indicate more or less protein than a lighter shade of blue?

 

 

5.  What is the purpose of the standards?

6.  What type of data will you collect with the spectrophotometer?

7.  Observe the cuvettes containing milk samples that have been mixed with Bradford dye reagent (2% = 2% milk, W = whole milk, Sk = skim milk, and So = soy milk).   Do they contain protein?  How can you tell?  If they contain protein, how will you quantify the actual amount of protein in each sample?

8.  In order to study the proteins found in the milk samples, you diluted the milk samples to 1 mg/mL and then loaded them in a vertical gel.

9.  Do the milk samples contain the same proteins?  How can you tell?
